I just got this new Endura and I find myself tending to open it with a quick flick of the thumb so it snaps open decisively, almost like an auto. Is opening the knife like this going to cause the lock to loosen up over time? I'm delighted with the way this knife performs so I don't want to abuse it. Any advice out there from long time Endura owners?

It will loosen but you can tighten it back up with a small torx wrench kit. I got a torx set off ebay for about 3$ and a very nice torx and small screwdriver set at home depot for about 8$

slamming the knife open as hard as you can will weaken the back spring. However a little thumb and a little wrist won't do any real damage to it.

By flicking your knife open with excessive, overtly "flashy" force, you will create premature wear on the lock interfaces. Open your knife with a smooth and controlled motion. If you want something with Assisted Opening or an auto, get one of those intead.
