I don't carry for self-defense, but I know that a lot of people consider (at least lightly) self defense with their EDC. People ignore SAK's as a self-defense tool, but I was thinking...
The blade is a slipjoint, so the best lock method would be to put your thum on the edge of the tang, and stab in a reverse grip with upward swing. But in a fight, that's probably not the best idea because you probably won't have your wits about you, and you could mess it up and hurt yourself. So the knife blades are out. BUT, what about the other tools? I think the best would be the:
1) Bottle opener. It locks open strong, and if it does shut it has the 90 degree stop and it's not a sharp edge. It would KILL to be hit with that thing concentrating the force, and you're going to get punctures. if it closes before you ward off an attacker, use the knife as a kubotan.You could also use the can opener, but I seem to think this works better.
2) Awl. This works with the rear-awl design, which you can wrap your fingers around and use to punch with. I put this second because it has slow deployment and if you have thick fingers you might not get the length you need.
3) Corkscrew. Faster deployment than the awl, and it would still hurt like crazy! This also works with phillips screwdriver (even the front one, I suppose).
Like I said, I wouldn't carry a self-defense knife, but many others would want one, so I think the SAK is a good option for someone in a low-risk area who doesn't want their EDC affected. I don't know if this was covered before, but I thought of it while I was wiping off my bottle opener and my finger slipped into the tip. Didn't hurt much, but with such little leverage it seemed proficient at causing pain. Also, it will look better in court than if you had a one handed opener and stabbed the attacker to death. The point with these is to deter rather than kill, just food for thought.
