I am looking at cleaning up a knife with plastic scales. They seem like nylon would be my best guess. Can I just work through the sanding grits like wood?

Yes. Go slow and do it by hand. Finish with a high grit around 1000-2000. Buff by hand with a plastic polish and a soft cloth.
