holes in folding knife scales

Holes definitely improve grip. They are the 4x4 of knife scale textures. Almost any knife handle could have improved grip by the addition of 1 or more holes.
Holes also aid when trying to manipulate the knife to open/close with one hand or with your weak side hand. The holes provide more anchor points for your fingers. Enabling you to grip the knife in multiple ways. The holes reduce weight and can be attractive.

Speed holes are most effective when you have to throw your knife for self defense while in a crosswind. :D

I have never noticed them bothering my grip at all. It’s not something I conciously feel myself gripping unless I focus on them.

I tend to dislike them because I feel they give too much access to the blade by other objects in my pocket.

this was another concern of mine. I dont keep other stuff in my knife pocket, but I did wonder about stuff getting in the holes, or the blade getting scratched up somehow.
 
Most of those knives are impractical to use except as desk accessories. I remember as a kid reading an Outdoor Life or Field & Stream article on field dressing Whitetail deer. The writer said he much preferred fixed blades because of how much easier they are to clean. Hell, a Griptillian has more nooks and crannies than an English muffin. You'd need a WaterPik to clean out dried blood from any of those knives.

So there's nothing in between desk orniment and cleaning a deer?

I you want to clean your modern folder you can always just take it apart it's not particularly difficult, but yes fixed blades are better for these chores.
 
A desk accessory is different than a desk ornament. At least with the former you can open an envelope. (It was a poor joke.)
