What's the purpose of this little screw that overlaps the locking spring part of the handle? I've seen it on several other flippers before, is it a way to vary how much retention the blade has when closed?
It is a lock bar stabilizer. It prevents the lockbar from being over extended in the opposite direction during unlocking the blade. This is to prevent tension loss in the lockbar.
