One of my favorite lines is from Spartan.

No, as I recall he was rather reluctant to part with it.
 
I don't see many movies, but the first line that pops into my mind is:

Best movie line ever to me came from Driving Miss Daisy when Dan Akroid said his farewells to his hired driver and stubborn mother when they made the trip to see her Uncle for a birthday party. "Good Luck, Good Bye, Good God." I could feel his pain and relief.
