Defensive knife, flipper or thumb studs?

Hole. I prefer having all four fingers wrapped around the handle for positive control.

I have older Benchmades with thumb holes and thumb studs. They are easy to open but not 100% foolproof because the holes/studs are small, but at least there is not a lot of blade detent to overcome during opening. I have a Kershaw Leek which is hard to open fast because the stud is very small and the flipper is also small and hard for my finger to find. I have a CRKT Delegate with assisted opening but it is hard to open quickly because of their built-in lock. I have a Kershaw Blur that is very easy to open because it has mild detent and big studs. It is probably my most reliable knife to open quickly, and my Para 2 is a close second. When I get better used to the Para 2 it will probably tie with the Blur. I'm considering adding a BM Stryker to my EDC, and wondering how reliable it will be for quick opening.

But seriously back to the original question- even better than a quick-opening folder would be a good pair of eyeballs to keep people from getting too close to you and a good pair of tennis shoes to get away from them.

If you're going to carry a blade specifically for self defense then you should carry a fixed blade. You depend on a folder in a self defense situation when it's what you happen to have in your pocket at the time. If you are going to carry a fixed blade for self defense then you'd be better off learning how to carry and use a pistol in a self defense situation. Carry both if you deen it necessary.

Personally, I carry a flipper usually because they are what I like and what I'm used to.
