Most Comfortable Handle..............

Folders: Spyderco Paramilitary 2 and Emerson CQC-10. (I'm on my second CQC-10 and it's the cat's meow).
Fixed: Busse Active Duty and Graham Razel. (Jon Graham makes some extremely ergonomic handles IMO).
 
Folder: Toss up between small sebbie, BM 615 minirukus, and spyderco mini manix (this would be the best if not for the aggressive G-10)
Fixed: BRKT mini fox river closely followed by my KA Swearingen Bird and trout
 
This one. No hot spots even with prolonged use. The smooth water buff horn scales are also surprisingly grippy even when wet.

Of folders the most comfortable are Kershaw Boa and BM 800 - 805 series. They are both of the greatests knifes in my opinion.
With fixed knives it is difficult to say. Those I have made myself I guess would be difficult to beat. :D
 
On a fixed blade, I think the Ratmandu from Swamp Rat knives is my most comfortable, but my Cold Steel Recon scout is a close second.
 
Nods to Becker, but I think Andy Roy (Fiddleback Forge) does some of the most comfortable handles in the business.
