The Kershaw Ken Onion Spec Bump is my most comfortable user. The 5" handle and contoured scales make it perfect for my hand.

Of folders the most comfortable are Kershaw Boa and BM 800 - 805 series. They are both of the greatests knifes in my opinion.
With fixed knives it is difficult to say. Those I have made myself I guess would be difficult to beat.

On a fixed blade, I think the Ratmandu from Swamp Rat knives is my most comfortable, but my Cold Steel Recon scout is a close second.
