I love a lot of Spydercos, and that is probably my favorite brand. My favorite blades at the moment are the Hinderer ZTs, but I'm taking a hard look at the Spyderco Southard, which is possibly the best blend of things I love in blades.
 
I voted thumb stud, thumb hole, and nail nick. Since my EDCs for daily use are paired fixed blades, I don't really care how the folders I carry open as long as it's a lock-back. Don't do slip joints or liner locks any more and don't like waves or flippers.
 
Probably the Spydie Hole.
But I'm cool with thumb studs/flippers.

Sorry, guys, I'm just not a fan of Spydercos.
 
In order:

1. Flipper
2. Spyderhole
3. Studs

And I agree with barres to a point. The Spyderhole looks strange and tends to add a lot of real estate to the blade. But the first time I used it I was hooked.
 
Flippers are easy and efficient, and wave, or something similar, my CRKT M21 has the same function but not the "wave shape" is very fast, but takes some practice, and can be intimidating, but very useful in a self defense moment, so I voted for the flipper and wave
