I have to say I love the flipper. Hole comes as a close second, but I just love the flipper. Gives the blade a much more attractive and sterile look, in my opinion, as well as being a simple, quick, and effective way of deployment.

Thumbstud and flipper would be my choice. I've always wanted Kershaw to make a Skyline XL with an inch longer handle/blade, quarter inch wider handle, larger thumbstud, keep the flipper, that would be a sweet knife. Make it FFG S35VN, boom.

Flipper/AO would be my preference with the "hole" a close second.

I say the "hole" because I have a Benchmade Grip with a hole and it opens like butter.

I like the ease of a flipper, albeit it I can deploy my Spydies just as fast. I just like them...why can't that be good enough? :p

Flipper hands down, but also has to have some sort of hole of stud for slow deployment.

I probably prefer the Spydie hole, as that is what my thumb is most used to. But a flipper is a close second. Though I like my Cold Steel products, their thumb plates are not my favorite.

Since no one has ever seen one of these before, this is what a Spydie hole looks like.

Really close for me between Spydie hole and flipper. I went with spydie hole because I find it a bit more pocket friendly. Flippers tend to get caught in my pocket or dig in uncomfortably when getting out keys, wallets, or the like. The Spydie hole also works on any size blade and any lock even if the blade is new or a bit stiff to open. That said, though, I love my knives with flippers for their convenience in opening. If my primary uses required fast one-hand opening on a regular basis I'd seriously consider the flipper over the spydie hole.

I voted for thumb studs just because that's what I'm most used to, but I don't having anything against any specific opening method. I have stuff with holes, studs, nicks, flippers, and even strange levers (CRKT Hole In One). As long as the blade is easy to deploy, I don't really care.
