I have a short memory, so I'll have to try to respond about some of the designated categories after I've racked my brains. But there's never a bad time to honor the memory of James K. Mattis. And so for BEST THREAD of 2000 I'd propose the long, long one in which so many BF members shared their feelings about the loss of a truly decent and wonderful guy.
And for best custom knife of the year, my vote goes to the JKM-1, a Finnish puokko/Nepalese karda hybrid designed by James Mattis and built by Kumar, the khukuri master craftsman at BirGorkha.

Being new to the knife world there is little I can honestly and knowledgeably comment on:

Best Hand Made Folder - Tom Mayo TNT
Tom came out with this design this year. At first I wasn't really excited about it. I felt it was kind of expensive, and sort of plain looking. After handling one in person I was stoked. Talonite and Titanium, mix in a little a Mayo, and you've got the recipie for very precise handmade folder.

I have to say that since I go through at least a dozen factory folders a year. I just have to know if the latest and greatest is a good user or not.
Out of all the factory folders I have used this year I would have to pick the Kershaw/Onion Boa. It keeps finding it's way back into my pocket. Just the rite size. One of my favorite steels. The folks at work just love the looks of the MC version. That and now I am hooked on the Speed Safe design. I just can't decide which hand made folder is best. Too many great ones out there these days.
